The Chengdu Institute of Biology, Chinese Academy of Sciences, was established in 1958. It is a research institute directly under the Chinese Academy of Sciences under the first-level discipline. Chengdu Institute of Biology Public Experimental Technology Center has a variety of common experimental equipment, with 600MHz nuclear magnetic, high-resolution mass spectrometry, amino acid automatic analyzer, multi-purpose microscope and other advanced equipment. At present, Chengdu Institute of Biology has achieved more than 300 scientific and technological achievements, of which more than 100 awards have been awarded by provincial and ministerial level scientific and technological achievements. All the time, Chengdu Biology Institute has been deeply involved in electrospray ionization technology.
Electrospray ionization technology was introduced in the 1970s. It has a soft ionization characteristic that does not cause the fragmentation of compounds. It is the most widely used ionization method in mass spectrometry. However, conventional techniques have the disadvantage of not directly analyzing high-salt biological samples. It is necessary to previously desalt high-salt samples or to use liquid chromatography using buffer salts.
In 2017, the development of research and development equipment for Chinese Academy of Sciences under the auspices of the Chengdu Institute of Biology, “Development of an Electrospray Ion Source for Biological Mass Spectrometer Probes†passed the final acceptance. Chengdu Institute of Biology has continuously optimized the use of liquids by optimizing the control method, sample loading method, high-voltage switch-on method, and ion transmission method to make it resistant to high-voltage interference, salt resistance, and matrix interference. The interface allows the ion source to be used in conjunction with liquid chromatography using a high-salt buffer solvent, and devices have been successfully developed.
In the process of research and development, the Chengdu Institute of Biology has encountered new problems. The electrospray ionization process is usually performed in a polar solvent. This ionization technique is suitable for ionization analysis of medium to high polarity systems. However, many compounds are only soluble in low-polarity solvents, and such samples are difficult to ionize by electrospray, thus making ESI-MS limited in the analysis of low-polarity solvent systems and the mechanism of partial organic reactions.
In response to the problems encountered, Chengdu Institute of Biological Science and Technology has proposed an electrospray mass spectrometer based on conductive nanomaterials and a method for electrospray mass spectrometry analysis. In addition to ionizing compounds dissolved in a polar solvent, the method can also ionize compounds dissolved in a low-polarity solvent, satisfy the mass spectrometry requirements of polar and low-polarity systems, and has a simple method. low cost.
